RP-HPLC determination of hyperoside in Burnet

【Abstract】 Objective To establish an HPLC method for the determination of hyperoside in Sanguisorba officinalis. Method RP-HPLC was used for determination, C18 column, mobile phase was methanol-0.2% phosphoric acid solution (46:54), pH was adjusted to 3.0 with triethylamine, flow rate was 1.0 ml · min-1, detection wavelength was 370 nm; for test product The solution was prepared using methanol ultrasonic extraction. Results The mass fractions of Sanguisorba officinalis from different origins were determined, and the mass fraction of hyperin was 0.052% to 0.291%. Conclusion The method is sensitive, fast, accurate and reliable, and can be used for the quality control of Burnet.

ConclusionThe method is accurate and can be used for the quality of Sanguisorba officinalis.

Sanguisorba officinalis L. or Sanguisorba officinalis L.var.longifolia (Bert.) Yu et Li's dried roots. Its taste is bitter, sour, astringent, slightly cold, and has the effect of cooling blood to stop bleeding, detoxification and sore [1, 2]. Modern pharmacological studies have shown that it has a significant analgesic effect [3], the main active ingredient is flavonoids, but the quantitative detection method described in the Pharmacopoeia quality standards is only ultraviolet-visible spectrophotometry to determine its tannin content, which is cumbersome to operate . Hypericin is a flavonoid compound extracted from Burnet. Studies have shown that hyperoside in Burnet is one of the effective ingredients for its analgesic effect. In this experiment, the RP-HPLC method was used to quantitatively analyze the composition of hypericin in the Chinese medicinal materials market from different places of origin, and the quality status of the commodities in the market was investigated, which provided a scientific basis for further improving the quality control of the Chinese medicinal materials. .
