ELISA kit decoding GPCR structure

It is not easy to analyze the structure of GPCR with ELISA kit. "We obtained the high-resolution structure of this important membrane protein for the first time at room temperature," said Vadim Cherezov of the Scripps Research Institute, who is the leader of this research. "At the same time, we have also verified a new technology that people can use to resolve new structures in the future."
Human serotonin receptors have been studied. The protein is involved in learning, mood and sleep, and is the target of obesity, depression and migraine drugs. To simulate the cellular environment, scientists prepared crystals of the receptor in fat gels. They used a new injection system to make the gel enter the path of LCLS (Linac Coherent Light Source) X-ray pulses to collect protein structure information.
This ELISA kit method solves the biggest obstacle in GPCR research: traditional X-ray studies using synchrotrons require larger crystals, which is difficult for GPCRs to achieve. Since LCLS is one million times brighter than the most powerful synchrotron and supports ultra-fast imaging, researchers can use this technique to analyze tiny crystals and collect data immediately. The LCLS imaging is too late to cause damage, so the sample does not need to be frozen and can be detected in a more natural state.
The human body has about 800 GPCRs, but so far, scientists have only analyzed more than a dozen GPCR structures. Previously, people have analyzed the structure of human serotonin receptors using traditional methods, but the conditions for conducting experiments at that time were not ideal. Now, the new technology of ELISA kit provides a more accurate serotonin receptor structure, which will help people better develop related drugs and avoid side effects.
